TECHNICAL DESCRIPTION
This invention describes a process to increase gene targeting frequency in E. coli (factor 1 000 to 100 000). Chemical modifications result in DNA damage, blocking of DNA replication and improvement of homologous recombination efficiency. This process allows gene targeting by integrating a modified gene in place of a healthy gene for the inactivation of said gene.
