ANTI-CANCER TREATMENT USING IMCs (Immature Myeloid Cells) AND REGULATORY T CELLS (Treg cells) AS TAR
Principal Investigator: Dr. Abdelilah WAKKACH

TECHNICAL DESCRIPTION

The accumulation of immature myeloid cells (IMCs) is often associated with a large tumor burden and a state of immune suppression characterized by an alteration of dendritic cells and T cell dysfunction. We have demonstrated that this state of immune suppression is associated with the amplification of Treg cells by IMCs and IL-10 appears to be the determining factor that acts as a costimulator for Treg cell amplification as well as inactivation of dendritic cells.

The invention consists of a gene therapy used for treating cancer, based on the increase of dendritic cells activity and inhibition of Treg cell amplification. The therapy is carried out by administrating a plasmid, expressing an active DNA enhancing immunostimulating activity.

This strategy has shown efficiency on 80 % of the tumors tested in vitro, and in vivo in mice.
