TECHNICAL DESCRIPTION
Starting from the observation that many ion-channels receptors such as GABA receptors are functionally and physically coupled to P2X receptors when they are co-expressed in a heterologous expression system, the inventors designed an original and convenient screening method for selecting or identifying modulators of said ion-channels receptors.
Based on a simple correlation between the calcium influx via the P2X receptor and the activity of the coupled ion-channel receptor, this technology provides a simple, rapid and reliable method to record, by measuring the calcium influx in co-transfected cells, the activity of the studied ion-channel receptor. Conveniently, fluorescent calcium probes can be used in High Throughput Screening.
